A Mysterious Case has been up since before the game was released, but Nalzen should give you a note that if you right click it, it will give you the quest to get you to that quest.
As for the tier 2 horse quest, it is actually dependent on finishing the tier 1 quest. The NPC will only give you the tier 1 horse if you have completed your tier 1 horse quest and gotten rid of your tier 1 horse.
